目的:观察局部冷冻对胶质瘤细胞凋亡的影响,以及不同冻融周期与细胞凋亡之间的关系,进一步探讨冷冻杀伤肿瘤细胞的分子机制。方法:建立胶质瘤动物模型,应用末端标记技术原位检测细胞凋亡, DNA琼脂糖电泳观察 DNA梯形条带。结果:经不同冻融周期冷冻 G422胶质瘤细胞后,在不同时间点,冷冻周围区的肿瘤细胞出现了形态学和 DNA水平上的细胞凋亡变化,其峰值出现在冷冻后 24~ 48 h, 重复冻融后出现更多的凋亡细胞。结论:冷冻治疗可以诱导 G422胶质瘤细胞发生凋亡,细胞凋亡的数目与冻融周期有关。诱导肿瘤细胞凋亡可能是冷冻杀伤肿瘤细胞的重要机制。
OBJECTIVE: To observe the effect of local freezing on the apoptosis of glioma cells and the relationship between different freeze-thaw cycles and apoptosis, and to further explore the molecular mechanism of freezing and killing tumor cells. Methods: Animal model of glioma was established. Apoptosis was detected by end labeling technique in situ. The DNA ladder was observed by DNA agarose gel electrophoresis. Results: After frozen-thawed G422 glioma cells at different time points, the apoptosis of the tumor cells in the surrounding frozen zone appeared morphological and DNA changes, the peak appeared in 24-48 h after freezing After repeated freeze-thaw, more apoptotic cells appeared. Conclusion: Cryotherapy can induce apoptosis in G422 glioma cells, and the number of apoptosis is related to the freeze-thaw cycle. Inducing apoptosis of tumor cells may be an important mechanism of freezing and killing tumor cells.
