采用纤维素酶酶解预处理与醇溶液浸提相结合的提取工艺从油松花粉中萃取黄酮类化合物,用NaNO2-Al(NO3)3-NaOH可见光分光光度显色法测总黄酮含量。探讨影响提取率的主要因素,并通过正交法确定最佳提取工艺。结果表明:各因素对提取效果影响的顺序依次为酶浓度>酶解时间>酶解温度>介质pH,确定最佳提取工艺为:酶浓度0.35g/L,酶解时间150min,酶解温度55℃,介质pH为5.0,最佳条件下不为例总黄酮得率达1.55%。
Flavonoids were extracted from the pollen of Pinus tabulaeformis by enzymolysis pretreatment combined with alcohol solution extraction. Total flavonoid content was measured by NaNO2-Al (NO3) 3-NaOH visible light spectrophotometry. The main factors influencing the extraction rate were explored, and the best extraction technology was determined by orthogonal test. The results showed that the order of influencing factors on the extraction was enzyme concentration> enzymolysis temperature> hydrolysis temperature> pH. The optimal extraction process was as follows: enzyme concentration 0.35 g / L, enzymolysis time 150 min, enzymolysis temperature 55 ℃, the pH of the medium is 5.0, under the optimal conditions, the yield of total flavonoids reached 1.55%.
