目的:探讨活血化瘀药物治疗类风湿性关节炎(RA)的机制。方法:以佐剂性关节炎(AA)大鼠为模型,观察血府逐瘀提取物对其血清肿瘤坏死因子-α(TNF-α)和一氧化氮(NO)及滑膜组织血管内皮生长因子(VEGF)表达的影响。结果:AA大鼠血清TNF-α和NO及滑膜组织中VEGF的表达均高于正常对照组;高、中剂量血府逐瘀提取物组的TNF-α和NO及滑膜组织中VEGF的表达低于模型组。结论:血府逐瘀提取物可改善AA大鼠足趾肿胀程度,降低病理评分,降低异常升高的TNF-α、NO及抑制滑膜组织中VEGF的过度表达,得出瘀血闭阻可能是RA的发病机制之一。
Objective: To investigate the mechanism of activating blood circulation and removing blood stasis drugs on rheumatoid arthritis (RA). Methods: The adjuvant arthritis (AA) rats were used as model to observe the effects of Xuefu Zhuyu extract on tumor necrosis factor-α (TNF-α), nitric oxide (NO) and synovial membrane vascular endothelial growth Factor (VEGF) expression. Results: The expression of TNF-α and NO in synovial tissue and the expression of VEGF in synovium of AA rats were higher than those in normal control group. The levels of TNF-α and NO in high-dose and middle-dose Xuefuzhuyu extract group and VEGF in synovial tissue Expression was lower than model group. Conclusion: Xuefu Zhuyu extract can improve the swelling degree of toes, reduce the pathological score, reduce the abnormally elevated TNF-α, NO and inhibit the over-expression of VEGF in synovium tissue, Is one of the pathogenesis of RA.
