目的:探讨细胞周期蛋白依赖性激酶2(Cdk2)和细胞周期蛋白E(CyclinE)在胆管细胞癌中的表达及临床意义。方法:收集武汉大学人民医院病理科2002-2008年胆管细胞癌存档蜡块40例,其中男性20例,女性20例。另取胆管细胞癌周围正常组织5例作对照。采用免疫组织化学方法检测各组中Cdk2蛋白和CyclinE的表达,利用HPIAS-2000图像分析系统测定各组中Cdk2蛋白和CyclinE表达的平均光密度和平均阳性面积率。结果:胆管细胞癌中Cdk2蛋白和CyclinE呈高表达,对照组中Cdk2蛋白和CyclinE呈低表达,胆管细胞癌组与对照组之间Cdk2蛋白和CyclinE表达的平均光密度及阳性面积率有显著性差异(P<0.05)。结论:Cdk2与CyclinE在胆管细胞癌的发生发展中发挥正性调节因子的作用。
Objective: To investigate the expression and clinical significance of Cdk2 and CyclinE in cholangiocarcinoma. Methods: Forty cases of paraffin-embedded paraffin wax were collected from Department of Pathology, People’s Hospital of Wuhan University from 2002 to 2008, including 20 males and 20 females. Another cholangiocellular carcinoma around the normal tissue 5 cases as a control. The expression of Cdk2 protein and CyclinE in each group were detected by immunohistochemical method. The average optical density and average positive area ratio of Cdk2 protein and CyclinE expression in each group were determined by HPIAS-2000 image analysis system. Results: The expression of Cdk2 protein and CyclinE in cholangiocarcinoma was high, while the expression of Cdk2 protein and CyclinE in cholangiocarcinoma was low. The average optical density and positive area ratio of Cdk2 protein and CyclinE in cholangiocarcinoma group and control group were significant Difference (P <0.05). Conclusion: Cdk2 and CyclinE play a positive regulatory role in the development of cholangiocarcinoma.
