用随机双脱氧核苷酸链末端终止法对人6型疱疹病毒(HHV-6)糖蛋白H(gH)基因进行DNA序列测定,找出开读框架,将其大部分插入到质粒pET3CP并分别转染大肠杆菌株BL21、PLYSS和PLYSE。35S甲硫氨基酸掺入试验表明,带有部分HHV-6gH基因片段的pET3CP质粒,可在上述大肠杆菌株表达,分子量约为37×10~3。
The DNA sequence of the glycoprotein H (gH) gene of human herpesvirus 6 (HHV-6) was determined by random dideoxynucleotide chain termination and the open reading frame was found, most of it was inserted into the plasmid pET3CP and respectively E. coli strains BL21, PLYSS and PLYSE were transfected. The 35S methionine amino acid incorporation assay showed that the pET3CP plasmid with a partial HHV-6gH gene fragment was expressed in the above E. coli strain with a molecular weight of about 37 × 10-3.
