目的 观察编码鼠 IL - 12的真核表达载体对 HBVDNA疫苗诱导 BAL B/ c小鼠免疫应答的影响 .方法 肌肉注射 DNA疫苗 ,EL ISA法检测小鼠血清抗 HBs,4h5 1 Cr释放法检测小鼠脾细胞 CTL杀伤活性 .结果 免疫 8wk后 ,单纯注射 p CR3.1- S及共注射 IL - 12真核表达载体的小鼠血清 45 0 nm A值分别为 0 .87± 0 .1及 1.6 7± 0 .15 .CTL 细胞杀伤活性分别为 (5 0 .5± 6 .4) %及 (73.3± 8.8) % ,两组均有明显差异 (P<0 .0 1) ,脾细胞悬液经抗 CD4+ 单克隆抗体处理后 CTL 细胞杀伤活性分别为 (48.3± 5 .9) % ,(75 . 6±9.1) % ,抗 CD8+单克隆抗体处理后分别为 (10 .6± 1.4) % ,(16 .9± 2 .3) % .结论 IL - 12的真核表达载体能够提高小鼠对 DNA疫苗的免疫应答 ,CTL细胞杀伤活性主要由 CD8+执行 .基因疫苗可能用于预防及治疗 HBV感染
Objective To observe the effect of eukaryotic expression vector encoding murine IL - 12 on the immune response induced by HBVDNA vaccine in BALB / c mice.Methods DNA vaccines were injected intramuscularly, the serum anti - HBs were detected by ELISA, the release of 4h5 1 Cr RESULTS: After 8 weeks of immunization, the 45 0 nm A values of mice injected with p CR3.1-S and co-injected with IL-12 eukaryotic expression vector were 0.87 ± 0.1 and 1.6 respectively 7 ± 0.15. CTL cell killing activity was (50.5 ± 6.4)% and (73.3 ± 8.8)%, respectively, with significant difference between the two groups (P <0.01) The cytotoxity of CTL cells treated by anti-CD4 + monoclonal antibody were (48.3 ± 5.9)% and (75.6 ± 9.1)%, respectively. The anti-CD8 + monoclonal antibody treatment was (10.6 ± 1.4) (16.9 ± 2.3)%, respectively.Conclusion The eukaryotic expression vector of IL - 12 can improve the immune response to DNA vaccine in mice and the CTL cell killing activity is mainly performed by CD8 + gene vaccine may be used to prevent and treat HBV infection
