目的 :为探讨神经膨体是否同时含有NOS和BDNF或BDNF和GDNF ,以间接了解背根节 (dorsalrootganglion ,DRG)小细胞是否同时含有这些物质 .方法 :用成年猫L3 脊髓节段制作 2 0 μm厚的冰冻切片并分两组 .一组先行NOS酶组化染色 ,NBT兰色反应显色 ;继之用BDNF抗血清以免疫组化ABC法行双标染色 ,DAB棕色反应显色 .另组先用BDNF抗血清以免疫组化ABC法染色 ,显色同前 ;随后用GDNF抗血清同前染片 ,TMB兰色反应显色 .常规脱水透明封片 .结果 :在NOS与BDNF双标组 ,脊髓Ⅱ板层内有大量NOS阳性 (呈兰色反应 )和BDNF阳性 (呈棕色反应 )神经膨体 ,但大多为单标染色 ,双标染色者极少 ;BDNF和GDNF双标组阳性神经膨体的标记与NOS和BDNF组相类似 ,均主要是大量单标的神经膨体 ,双标者少 .结论 :脊髓Ⅱ板层虽有NOS ,BDNF和GDNF阳性神经膨体 ,但各自来源有所不同 ,即大多数DRG小细胞并非同时含有NOS和BDNF或BDNF和GDNF .提示以上 3种阳性神经膨体可能分别来源于不同功能类型的DRG小细胞
OBJECTIVE: To investigate whether NGF and BDNF or BDNF and GDNF are involved in the development of neurofibers in order to find out whether dorsal root ganglion (DRG) small cells contain these substances at the same time.Methods: Adult adult cat L3 spinal cord segments were used to make 20 μm Thick frozen slices and divided into two groups.A group of first NOS enzyme staining, NBT blue reaction color; followed by BDNF antiserum by double staining immunohistochemical ABC method, DAB brown reaction color. The other group First with BDNF antiserum by immunohistochemical ABC staining, color with the former; followed by GDNF antiserum with the former dye, TMB blue color reaction routine dehydration clear packing.Results: NOS and BDNF double standard group There were a large number of NOS positive (blue reaction) and BDNF positive (brown reaction) neuroplasticity in the spinal cord II platelet, but most of them were single-labeled and few double-stained ones. BDNF and GDNF double positive neurons Compared with NOS group and BDNF group, the bulge markers were mainly a large number of single-labeled neurite and few double markers.Conclusion: Although there are NOS, BDNF and GDNF-positive neuroplastic bulimia in the lamina Ⅱ, In contrast, most DRG minicells do not contain both NOS and BDNF or BDNF And GDNF.It is suggested that the above three types of positive nerve bulge may originate from different functional types of DRG minicell
