目的:探讨醋柴胡不同提取部位治疗肝郁证的效果及可能机制。方法:SD大鼠随机分出空白组,其余夹尾制备肝郁模型。造模成功后给予醋柴胡不同部位提取液治疗3d,测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、总胆酸(TBA),肝脏组织的Na+-K+ATP酶、Ca2+-Mg2+ATP酶活性,RT-PCR法检测P-gp m RNA含量。结果:造模后,与空白组比较,模型组各肝功检测指标均升高(P<0.05,P<0.01),而醋柴胡多个部位均能降低肝郁模型大鼠的ALT、AST和TBA水平;增强ATP酶活力,其中水提液部位(WE)、正丁醇部位(BE)的Na+-K+、Ca2+-Mg2+ATP酶活力明显提高(P<0.05);RT-PCR实验显示醋柴胡各部位均能降低P-gp基因表达(P<0.05,P<0.01)。结论:醋柴胡各部位能够改善肝郁证导致的肝损伤,其机制与Na+-K+ATP酶、Ca2+-Mg2+ATP酶、P-gp调控有关。
Objective: To investigate the effect and possible mechanism of different extracts of vinegar on Bupleurum chinense. Methods: SD rats were randomly divided into blank group, the rest of the clip tail preparation of liver depression model. After the model was established, the extracts of different parts of vinegar were given for 3 days. Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholic acid (TBA), hepatic Na + -K + ATPase, Ca2 + -Mg2 + ATPase activity, P-gp m RNA content was detected by RT-PCR. Results: Compared with the blank group, all indexes of liver function in the model group were significantly increased (P <0.05, P <0.01), while multiple sites of vinegar could reduce the ALT, AST (P <0.05). The results of reverse transcription polymerase chain reaction (RT-PCR) showed that the activities of ATPase, All parts of vinegar could reduce the expression of P-gp gene (P <0.05, P <0.01). Conclusion: All parts of vinegar can improve hepatic injury induced by liver depression. The mechanism is related to the regulation of Na + -K + ATPase, Ca2 + -Mg2 + ATPase and P-gp.
