壳聚糖及其衍生物包覆对载姜黄素脂质体角膜透过作用的影响

采用乙醇注入法制备姜黄素脂质体,并分别以壳聚糖、壳寡糖、三甲基壳聚糖(TMC)及羧甲基壳聚糖包覆脂质体。采用离体兔角膜透过试验及共聚焦激光扫描显微镜成像技术(CLSM),考察壳聚糖及其衍生物包覆的脂质体对角膜透过作用的影响。结果显示,TMC包覆脂质体的促角膜渗透作用最强;且随作用时间的延长,脂质体可渗透至距角膜上皮层60μm。CLSM成像结果显示TMC包覆脂质体主要以细胞旁路途径透过角膜。与未包覆脂质体相比,TMC包覆脂质体透过离体角膜的稳态流量显著增加,且表观渗透系数提高。 The curcumin liposomes were prepared by ethanol injection, and the liposomes were coated with chitosan, chitooligosaccharides, trimethylchitosan (TMC) and carboxymethyl chitosan. In vitro rabbit corneal permeability test and confocal laser scanning microscopy (CLSM) were used to investigate the effects of liposomes coated with chitosan and its derivatives on corneal permeability. The results showed that the TMC-coated liposomes had the strongest corneal permeation effect, and the liposomes could penetrate 60μm away from the corneal epithelium with the extension of time. CLSM imaging results show that TMC-coated liposomes mainly through the cornea by the cell-bypass pathway. Compared with uncoated liposomes, the steady state flow of TMC-coated liposomes through corneal exfoliation increased significantly, and the apparent permeability coefficient increased.
