目的 研究肝肾宝对 5 / 6肾切除大鼠肾组织细胞外基质 (ECM)如纤维粘连蛋白 (FN)、层粘连蛋白 (LN)、Ⅳ型胶原 (ColⅣ )蛋白及其基因表达的影响。方法 采用免疫组化、原位杂交方法检测肾组织FN、LN、ColⅣ及相应mRNA表达 ,使用TN - 85 0 2多功能图像分析系统对结果进行处理 ,采用SPSS统计软件统计。结果 显示正常对照组、假手术组 ,均未见FN、LN、ColⅣ蛋白及其mRNA阳性反应物。 5 / 6NT组 ,肾小球细胞细胞浆、肾小管细胞浆见FN、LN、ColⅣ蛋白及其mRNA强阳性黄色反应物。缬沙坦组FN、LN、ColⅣ蛋白及其mRNA阳性反应物减少 ,与 5 / 6NT组比较 ,P <0 0 5 ;肝肾宝组FN、LN、ColⅣ蛋白及其mRNA阳性反应物明显减少 ,与 5 / 6NT组比较 ,P <0 0 1。结论 肝肾宝下调肾组织FN、LN、ColⅣ蛋白及其基因表达 ,是防治肾纤维化作用机制之一
Objective To investigate the effects of Ganshen Shenbao on the expression of extracellular matrix (ECM) such as fibronectin (FN), laminin (LN), type IV collagen (ColIV) protein and its gene expression in 5/6 nephrectomized rats. Methods Immunohistochemistry and in situ hybridization were used to detect the expression of FN, LN, Col IV and corresponding mRNA in renal tissue. The results were processed using the TN-8502 multi-function image analysis system and statistical analysis was performed using SPSS statistical software. The results showed that normal control group, sham group, no FN, LN, ColIV protein and its mRNA positive reactants. In the 5/6NT group, glomerular cell cytoplasm and tubular cytoplasm were found to be strongly positive yellow FN, LN, Col IV proteins and mRNA. In valsartan group, FN, LN, ColIV protein and mRNA expression were decreased. Compared with 5/6NT group, P<0.05; Hepatospinal group significantly decreased FN, LN, Col IV protein and mRNA mRNA expression. Compared with 5 / 6NT group, P <0 0 1. Conclusion Liver-Kidney Poor Down-regulation of FN, LN, Col IV protein and its gene expression in renal tissue is one of the mechanisms of prevention and treatment of renal fibrosis
