目的:探讨石榴多酚对自发性高血压大鼠(SHR)的降压作用及其可能的作用机制。方法:40只12周龄雄性SHR大鼠随机分为5组:SHR对照组,石榴多酚低、中、高剂量组(120,240,480 mg·kg~(-1))和卡托普利(15 mg·kg~(-1))组。16只Wistar-Kyoto(WKY)大鼠随机分为2组:WKY对照组和石榴多酚480 mg·kg~(-1)组,连续灌胃给药4周。每周检测收缩压(SBP),舒张压(DBP),给药4周后,动物禁食12 h,25%乌拉坦麻醉,腹主动脉取全血。酶联免疫法检测大鼠血浆血管紧张素Ⅱ(AngⅡ)和内皮素(ET),硝酸还原法检测血清一氧化氮(NO)含量。结果:石榴多酚240,480 mg·kg~(-1)能显著降低SHR大鼠SBP、DBP和血清Ang II和ET含量,显著升高血清NO含量(P<0.05或P<0.01)。石榴多酚对WKY大鼠血压及血管活性物质含量无明显影响。结论:石榴多酚能够显著降低SHR大鼠血压,其机制可能与调节血管活性物质Ang II、ET、NO表达有关。
Objective: To investigate the antihypertensive effect of pomegranate polyphenols on spontaneously hypertensive rats (SHR) and its possible mechanism. Methods: Forty male SHR rats, aged 12 weeks, were randomly divided into five groups: SHR control group, low, medium and high doses of pomegranate polyphenols (120,240,480 mg · kg -1) and captopril · Kg ~ (-1)) group. Sixteen Wistar-Kyoto (WKY) rats were randomly divided into two groups: WKY control group and pomegranate polyphenols 480 mg · kg -1 group. Systolic blood pressure (SBP) and diastolic blood pressure (DBP) were measured weekly. After 4 weeks of administration, the animals were fasted for 12 hours, 25% of urethane anesthesia and whole abdominal aorta were taken. Plasma levels of angiotensin Ⅱ (Ang Ⅱ) and endothelin (ET) were measured by enzyme-linked immunosorbent assay (ELISA), and serum nitric oxide (NO) was measured by nitrate reduction. Results: Pomegranate polyphenol 240,480 mg · kg -1 could significantly decrease SBP, DBP, Ang II and ET in serum and NO in serum (P <0.05 or P <0.01). Pomegranate polyphenols on WKY rats blood pressure and vasoactive substance content had no significant effect. Conclusion: Pomegranate polyphenols can significantly decrease the blood pressure in SHR rats, which may be related to the regulation of the expression of Ang II, ET and NO.
