目的研究氯喹(chloroquine,CQ)对杨梅素诱导的He La细胞凋亡的影响。方法体外培养宫颈癌He La细胞,实验分为Control组、单加杨梅素(50μg/ml)组、单加CQ(20μM/ml)组、合加药物组(杨梅素50μg/ml+CQ 20μg/ml)共4组,观察药物对He La细胞增殖率的影响。共聚焦显微镜下观察He La细胞核的形态变化、凋亡相关蛋白及自噬标志物的表达。结果与单加杨梅素组相比,合加药物组He La细胞增殖率明显下降,凋亡形态学改变更加明显及凋亡相关蛋白active caspase-3表达增加;自噬标志物微管相关蛋白轻链3(microtubule-associated protein light chain 3,LC3)表达增加。结论氯喹能够促进杨梅素诱导的HeLa细胞凋亡。
Objective To investigate the effect of chloroquine (CQ) on the apoptosis of Hela cells induced by myricetin. Methods HeLa cells were cultured in vitro. The experiment was divided into three groups: Control group, single myricetin (50μg / ml) group and CQ alone (20μM / ml) ml) for 4 groups. The effects of drugs on the proliferation rate of He La cells were observed. Confocal microscopy was used to observe the morphological changes of He La nuclei and the expression of apoptosis-related proteins and autophagy markers. Results Compared with the single myricetin group, the proliferation rate of He La cells was significantly decreased, the morphological changes of apoptosis were more obvious and the expression of active caspase-3 was increased. The expression of autophagy marker microtubule-associated protein Increased expression of microtubule-associated protein light chain 3 (LC3). Conclusion Chloroquine can promote myricetin-induced apoptosis in HeLa cells.
