目的观察滋阴泻火方对环境内分泌干扰物壬基酚(NP)及双酚A(BPA)的拟雌激素活性的拮抗作用。方法30只3周龄雌性SD大鼠随机分为对照组(喂饲玉米油),染毒A组(喂饲NP100mg/kg),染毒B组(喂饲NP50mg/kg+BPA200mg/kg),治疗A组(喂饲NP100mg/kg+滋阴泻火方),治疗B组(喂饲NP50mg/kg+BPA200mg/kg+滋阴泻火方),疗程15天,检测子宫增重试验及增殖细胞核抗原(PCNA)蛋白表达。结果染毒A、B组与对照组比较子宫湿重、子宫脏器系数、子宫内膜及平滑肌厚度、子宫内膜腺体及腺上皮高度显著增加(P<0.05),子宫内膜及肌层的PCNA蛋白表达显著增高(P<0.05)。治疗A、B组分别与染毒A、B组相比,上述各指标均明显降低(P<0.05)。结论壬基酚及双酚A具有显著的拟雌激素活性,滋肾阴泻相火中药对壬基酚及双酚A的拟雌激素活性具有显著的拮抗作用。
Objective To observe the antagonism of Ziyinxiexiefang on the estrogenic activity of environmental endocrine disruptors, nonylphenol (NP) and bisphenol A (BPA). Methods Thirty female Sprague Dawley rats aged 3 weeks were randomly divided into control group (feeding corn oil), group A (feeding NP100mg/kg), and group B (feeding NP50mg/kg+BPA200mg/kg). Treatment group A (feeding NP100mg/kg + Ziyin Xiehuo side), treatment group B (feeding NP50mg/kg + BPA 200mg/kg + Ziyin Xiehuo side), treatment for 15 days, test uterine weight gain test and proliferating cell nuclear antigen ( PCNA) protein expression. Results The uterine wet weight, uterine organ coefficient, endometrial and smooth muscle thickness, endometrial glandular and glandular epithelial height were significantly increased (P<0.05), endometrium and myometrium were compared between A and B groups and control groups. The expression of PCNA protein was significantly increased (P<0.05). In the treatment group A and B, compared with the poisoned groups A and B, the above indexes were significantly lower (P<0.05). Conclusion Nonylphenol and bisphenol A have significant estrogenic activity. The traditional Chinese medicine with nourishing kidney and diarrhea has significant antagonistic effect on the estrogenic activity of nonylphenol and bisphenol A.
