目的探讨银杏叶提取物对支气管哮喘大鼠气道重塑的影响。方法 30只雌性SD大鼠,随机分为三组(每组10只):对照组、哮喘组和治疗组。采用卵蛋白腹腔注射和雾化吸入激发法复制哮喘大鼠模型。在末次激发24h后取动物肺组织行HE染色和Mas-son观察病理学改变;免疫组化测定转化生长因子β1(TGF-β1)表达。结果与对照组比较,哮喘组大鼠气道纤维化明显,TGF-β1表达增高(P<0.05);与哮喘组比较,银杏叶提取物治疗组中大鼠气道纤维化程度及TGF-β1表达减轻(P<0.05)。结论银杏叶提取物可抑制哮喘大鼠气道重塑的发生,其机制有可能是通过抑制TGF-β1的表达而实现的。
Objective To investigate the effects of Ginkgo biloba extract on airway remodeling in asthmatic rats. Methods Thirty female Sprague-Dawley rats were randomly divided into three groups (n = 10): control group, asthma group and treatment group. The asthmatic rat model was reproduced by intraperitoneal injection of ovalbumin and by inhalation stimulation. 24h after the last challenge animals were taken from the lung tissue HE staining and Mas-son pathological changes were observed; immunohistochemical determination of transforming growth factor β1 (TGF-β1) expression. Results Compared with the control group, the airway fibrosis and the expression of TGF-β1 in the asthma group were significantly increased (P <0.05). Compared with the asthma group, the degree of airway fibrosis and the expression of TGF-β1 in the Ginkgo biloba extract- The expression was reduced (P <0.05). Conclusion Ginkgo biloba extract can inhibit the airway remodeling in asthmatic rats, and its mechanism may be through inhibiting the expression of TGF-β1.
