目的 :探讨不同剂量、不同溶剂的基因重组技术生产的组织型纤溶酶原激活物 (r tPA) ,对于不同脱离时间的视网膜毒性作用。方法 :采用经外路显微手术方法 ,将Healon注入脉络膜视网膜下腔 ,制作出 30只脉络膜视网膜脱离兔眼模型 ;将不同剂量、不同溶剂的r tPA注入不同网脱天数的玻璃体腔内 ;用ERG、光学显微镜、电子显微镜等方法测定实验效应 ;用正交设计方法分析实验结果。结果 :脱离的视网膜对r tPA的毒性耐量明显降低 ,且脱离的时间越长 ,视网膜对r tPA耐量越低 (P <0 .0 1) ;不同的溶剂亦对这种耐受性有较明显的影响 (P <0 .0 5 )。结论 :脱离状态的视网膜对r tPA的毒性耐量降低 ,临床应用中应予注意
Objective: To investigate the effects of tissue plasminogen activator (r tPA) produced by gene recombination technology of different dosages and different solvents on retinal toxicity at different detachment time. Methods: 30 cases of choroidal retinal detachment rabbit model were made by injecting Healon into the choroid subretinal space by external microsurgery. The r tPA of different doses and different solvents were injected into the vitreous cavity with different days of cyst dendrites. ERG, optical microscopy, electron microscopy and other methods to determine the experimental effect; orthogonal design method to analyze the experimental results. Results: Detachment of the retina significantly reduced the toxic tolerance of r tPA, and the longer the detachment, the lower the tolerance of the retina to r tPA (P <0.01); different solvents were also more tolerant of this (P <0.05). Conclusion: Detached retinal damage to r tPA reduced toxicity, should pay attention to clinical application
