目的 :为了分离、培养肾脏内髓集合管 (IMCD)细胞。方法 :本文采用胶原酶消化和低渗溶解等步骤分离家兔肾脏加IMCD细胞 ;分离的细胞置DMEM /F1 2培养基、37℃、5 %CO2 的空气环境中培养 ;经透射电镜和角蛋白抗体免疫细胞化学等鉴定。结果 :几乎所有的培养细胞均被染为棕黄色 ;电镜观察培养的细胞核大 ,呈圆形或椭圆形。胞浆少 ,细胞器少见。细胞膜表面存在许多短而粗的微绒毛。与天然的IMCD细胞的形态和免疫细胞化学特征相符。结论 :本文培养的细胞为家兔肾脏IMCD细胞。
Objective: To isolate and culture renal medullary collecting duct (IMCD) cells. Methods: Rabbit kidneys plus IMCD cells were isolated by collagenase digestion and hypotonic lysis. The isolated cells were cultured in air at 37 ℃ and 5% CO2 with DMEM / F1 2 medium. The cells were cultured under TEM and keratin Antibody immunocytochemistry and other identification. Results: Almost all of the cultured cells were stained brown. Electron microscopy showed that the cultured nuclei were large and round or oval. Less cytoplasm, rare organelles. There are many short and thick microvilli on the cell membrane surface. Consistent with the morphological and immunocytochemical features of native IMCD cells. Conclusion: The cells cultured in this study are rabbit kidney IMCD cells.
