应用琼脂扩散沉淀试验(AGP)和微量血凝抑制试验(HI),对同胚接种鸡传染性支气管炎(IB)-新城疫(ND)二联苗的传染性支气管炎免疫效果进行监测。经人工感染IBV的鸡胚尿囊液,通过用蔗糖10-20倍浓缩后制作的IB-AGP抗原进行对比试验,结果表明IB-AGP可用作为IB的抗体的定性监测。经上法浓缩后的尿囊液,用等体积4%胰酶37℃处理2.5小时后,再用经56℃30分钟灭活的犊牛血清来终止反应,以此可制得较高滴度的IB-HI抗原。试验表明,IBV-HI具有特异性且其敏感性较高。两者比较,当IBV-HI价达6log2以上时IBV-AGP呈阳性,而且HI价越高AGP沉淀线也越明显。本试验结果表明:IBV-HI试验具有简单易行、准确快速、特异性和敏感性较高等优点,是监测鸡血清中IB抗体(Ab)的有效方法,并适宜于在基层单位和较大规模鸡场推广普及。而IBV-AGP虽然也简便易行,然而因其敏感度较低,若作为IB抗体的监测方法推广尚需改进和探索。
The effect of inoculation bronchitis (IB) - Newcastle disease (ND) vaccine inoculated bronchitis of invader was monitored by agar diffusion precipitation assay (AGP) and microagglutination inhibition test (HI). The IBV AGV was infected by human IBV AGV antigen and compared with IB-AGP antigen prepared by 10-20 times sucrose concentration. The results showed IB-AGP could be used as a qualitative monitor for IB antibody. The allantoic fluid concentrated in the above method was treated with an equal volume of 4% trypsin at 37 ° C for 2.5 hours and then stopped with calf serum inactivated at 56 ° C for 30 minutes, whereby a higher titer IB-HI antigen. Experiments show that, IBV-HI is specific and its sensitivity is high. When the price of IBV-HI was more than 6log2, IBV-AGP was positive, and the higher the HI value, the more obvious the AGP precipitation line. The results showed that: IBV-HI test has the advantages of simple and easy, accurate and rapid, high specificity and sensitivity, it is an effective method to monitor IB antibody (Ab) in chicken serum, and is suitable for the grassroots units and large-scale Chicken farms to promote universal. Although IBV-AGP is also simple and convenient, its sensitivity is low. Therefore, it is still necessary to improve and explore IBV-AGP as a monitoring method of IB antibody.
