用DNA条形码技术对市售花胶进行基原鉴定

目的:运用DNA条形码技术确定广州市售花胶的物种来源。方法:用自行设计的COⅠ引物对11份市售花胶样品进行PCR扩增、测序,将所得序列构建UPGMA系统发育树,进行聚类分析;用BOLD(The Barcode of Life Data Systems)系统的物种鉴定引擎,与数据库中已有鱼类序列进行比对,鉴定出各样品的基原。结果:11份市售花胶样品用自行设计的引物均可实现扩增,条带清晰、稳定,扩增和测序成功率均为100%;BOLD鉴定结果显示,11份样品来源于3科5种鱼类,UPGMA聚类分析对结果进行了确证。结论:DNA条形码技术与BOLD鉴定系统相结合,可对市售花胶进行准确的基原鉴定。 OBJECTIVE: To determine the origin of species in Guangzhou by using DNA barcode technology. Methods: 11 samples of commercial guar gum were amplified by PCR using self-designed COⅠ primers. The obtained sequences were used to construct the phylogenetic tree of UPGMA for cluster analysis. The BOLD (Barcode of Life Data Systems) The identification engine compares the existing fish sequences in the database to identify the bases of each sample. Results: 11 commercially available samples of gelatin could be amplified with primers designed by themselves. The bands were clear and stable, and the successful rate of amplification and sequencing was 100%. The BOLD results showed that 11 samples came from 3 families Species of fish, UPGMA cluster analysis confirmed the results. Conclusion: The combination of DNA barcode technology and BOLD identification system can accurately identify the commercially available gums.
