采用高碘酸钠氧化法首次合成酸枣仁皂苷A(jujuboside A)人工抗原,利用基质辅助激光解析电离-飞行时间质谱法(MALDI-TOF-MS)鉴定酸枣仁皂苷A人工抗原偶联成功,通过酶联免疫吸附测定法(enzyme-linked immunosorbent assay,ELISA)检测免疫小鼠血清中抗酸枣仁皂苷A抗体效价及其特异性。竞争抑制曲线表明,血清中的抗体可与酸枣仁皂苷A产生特异性结合,血清抗体效价在1∶4 000。成功合成的酸枣仁皂苷A人工抗原,为酸枣仁皂苷A单克隆抗体的制备及研究酸枣仁皂苷A在机体内的药物代谢过程奠定了基础。
The jujuboside A artificial antigen was synthesized for the first time by the sodium periodate oxidation method and the jujuboside A artificial antigen was successfully identified by matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF-MS) Anti-Jujuboside A antibody titer and specificity in serum of immunized mice were detected by enzyme-linked immunosorbent assay (ELISA). The competitive inhibition curve shows that antibodies in serum can specifically bind to Jujuboside A with a serum antibody titer of 1: 4000. The successful synthesis of jujuboside A artificial antigen, jujuboside A monoclonal antibody preparation and study Jujuboside A in the body of the drug metabolism process laid the foundation.
