为了测定阿奇霉素的含量,试验采用高效液相色谱(HPLC)法,以Kromasil C_(18)为色谱柱(250.0 mm×4.6 mm,5.0μm),以0.067 mol/L磷酸二氢铵(三乙胺调p H值为6.5)︰乙腈=70︰30为流动相,流速为1.0 m L/min,用UV230+型紫外-可见波长检测器,检测波长为210 nm,用外标法定量。结果表明:阿奇霉素在1.4~3.4 mg/m L的浓度范围内线性关系良好(r=0.999 8,n=6);样品的平均回收率为100.20%,相对标准偏差(RSD)=0.38%(n=3);样品测定方法重现性的RSD=0.49%(n=6),说明该方法可用于阿奇霉素含量的测定。
In order to determine the azithromycin content, the high performance liquid chromatography (HPLC) method was used on the Kromasil C 18 column (250.0 mm × 4.6 mm, 5.0 μm) with 0.067 mol / L ammonium dihydrogen phosphate (triethylamine Adjusted p H value of 6.5): acetonitrile = 70: 30 as the mobile phase at a flow rate of 1.0 m L / min. UV230 + UV-Vis detector was used. The detection wavelength was 210 nm and was quantified by external standard method. The results showed that azithromycin had a good linearity (r = 0.999 8, n = 6) in the range of 1.4-3.4 mg / m L; the average recovery was 100.20% and the relative standard deviation was 0.38% = 3). The RSD of reproducibility was 0.49% (n = 6), which indicated that this method could be used for the determination of azithromycin.
