目的探讨细胞间隙连接蛋白(connexin,Cx)43在良、恶性前列腺组织中的表达和分布情况,以及与整合素连接激酶(Integrin-linkedKinase,ILK)表达的相互关系。方法采用免疫组织化学SP法检测Cx43、ILK在7例正常前列腺组织(NP)、16例BPH及52例前列腺癌(Pca)组织中的表达。结果Cx43既定位于细胞膜上,又在细胞质中表达。Cx43的阳性表达率在Pca组织中为32.7%(17/52)显著低于NP组织的85.7%(6/7)、BPH组织的87.5%(14/16)(P<0.01;P<0.005)。Cx43在Pca细胞中的阳性表达率与肿瘤细胞的分化程度呈正相关(P<0.05),C+D期显著低于A+B期(P<0.05)。Cx43阳性表达与ILK的表达呈负相关(P<0.001)。结论Cx43异常表达于BPH、Pca细胞的胞浆。Cx43表达的增加或下降在BPH的增殖过程中或Pca的分化、进展中起重要作用。Cx43和ILK蛋白在Pca组织中呈现相反的的表达方式,提示在Pca细胞增殖动力学调节中协同促进了肿瘤的恶性进展。
Objective To investigate the expression and distribution of connexin (Cx) 43 in benign and malignant prostatic tissues and its relationship with the expression of integrin-linked kinase (ILK). Methods Immunohistochemical SP method was used to detect the expression of Cx43 and ILK in 7 cases of normal prostate tissue (NP), 16 cases of BPH and 52 cases of prostate cancer (Pca). Results Cx43 was localized both in the cell membrane and in the cytoplasm. The positive expression rate of Cx43 in Pca tissue was 32.7% (17/52), which was significantly lower than that in NP tissue (85.7%, 6/7) and 87.5% (14/16) in BPH tissue (P <0.01; P <0.005) . The positive expression rate of Cx43 in Pca cells was positively correlated with the differentiation degree of tumor cells (P <0.05), while the C + D phase was significantly lower than that of A + B phase (P <0.05). The positive expression of Cx43 was negatively correlated with the expression of ILK (P <0.001). Conclusion Cx43 is abnormally expressed in the cytoplasm of BPH and Pca cells. The increase or decrease of Cx43 expression plays an important role in the proliferation of BPH or the differentiation and progression of Pca. The expression of Cx43 and ILK protein in Pca tissue was reversed, suggesting that synergistic promotion of the malignant tumor progression in Pca cell proliferation kinetics.
