目的探讨非小细胞型肺癌组织中CUG结合蛋白1(CUGBP1)mRNA及蛋白的表达以及与预后的关系。方法选取2009年7月至2011年4月在青岛大学附属医院胸外科接受手术治疗的57例非小细胞型肺癌患者,男32例,女25例;年龄43~74(60.6±8.9)岁。通过半定量逆转录-聚合酶链反应(RT-PCR)及免疫组织化学技术检测癌组织及周围正常组织CUGBP1的基因和蛋白表达情况。所有患者术后每2个月电话随访1次,采用肿瘤进展时间(TTP)作为观察评估指标,分析CUGBP1 mRNA及临床病理各指标与预后的关系。采用χ2检验对CUGBP1mRNA及蛋白的表达进行统计分析,采用COX单因素及多因素分析CUGBP1 mRNA对于肺癌预后的意义。结果 CUGBP1 mRNA及CUGBP1蛋白在肺癌组织中高表达,并且与TNM分期及分化密切相关。通过单因素及多因素分析,CUGBP1 mRNA[P=0.007 4,HR=3.701,95%CI(1.420,9.648)]、TNM分期[P<0.000 1,HR=4.043,95%CI(2.098,7.794)]及年龄(P=0.001 8,HR=3.207,95%CI(1.544,6.664)]可以作为非小细胞肺癌患者的独立预后因素。结论 CUGBP1 mRNA及蛋白在肺癌组织中高表达。作为非小细胞肺癌患者的独立预后因素,CUGBP1 mRNA高表达患者预后较差。
Objective To investigate the expression of CUG binding protein 1 (CUGBP1) mRNA and protein in non-small cell lung cancer and its relationship with prognosis. Methods From July 2009 to April 2011, 57 patients with non-small cell lung cancer undergoing surgery in Department of Thoracic Surgery, Affiliated Hospital of Qingdao University were enrolled. There were 32 males and 25 females, ranging in age from 43 to 74 (60.6 ± 8.9) years. The gene and protein expression of CUGBP1 in cancer tissues and surrounding normal tissues were detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry. All patients were followed up every two months by telephone and the time to tumor progression (TTP) was taken as the evaluation index to analyze the relationship between CUGBP1 mRNA and clinicopathological parameters and prognosis. The χ2 test was used to analyze the expression of CUGBP1 mRNA and protein. The significance of CUGBP1 mRNA expression in prognosis of lung cancer was analyzed by COX univariate and multivariate analysis. Results CUGBP1 mRNA and CUGBP1 protein were highly expressed in lung cancer tissues, and were closely related to TNM staging and differentiation. According to univariate and multivariate analysis, CUGBP1 mRNA [P = 0.007 4, HR = 3.701,95% CI (1.420,9.648)], TNM stage [P 0.0001, HR 4.043, 95% CI 2.098, 7.794) ] And age (P = 0.001 8, HR = 3.207,95% CI (1.544,6.664)] could be used as independent prognostic factors in patients with non-small cell lung cancer.Conclusion CUGBP1 mRNA and protein are highly expressed in lung cancer tissues.As a non-small cell lung cancer Patients with independent prognostic factors, CUGBP1 mRNA expression in patients with poor prognosis.
