目的优选AB-8大孔树脂纯化苁蓉总苷的放大工艺,确定生产工艺参数。方法以松果菊苷、苁蓉总苷含量为指标,考察树脂种类、用量、吸附次数、洗脱剂乙醇浓度、用量对工艺的影响,确定最佳工艺条件,并通过中试及生产验证优选的纯化工艺。结果纯化工艺采用AB-8大孔树脂分离纯化,药材-树脂比为1∶1,上样吸附两次,分别用4倍量、3倍量水洗脱除杂,用40%乙醇5倍量、4倍量洗脱,收集洗脱液。结论该工艺简单可行、分离效果好,适用于苁蓉总苷的连续工业化生产。
Objective Optimization of AB-8 macroporous resin purification Cistanche glycosides amplification process to determine the production process parameters. Methods The contents of echinacoside and cistanchis glycoside were taken as indexes to study the effects of resin type, dosage, adsorption times, eluent ethanol concentration and dosage on the process, and the optimum process conditions were determined. Through the pilot plant and production verification Purification process. Results The purification process was separated and purified by AB-8 macroporous resin. The ratio of drug-resin was 1: 1, and the sample was adsorbed twice. The samples were eluted with 4-fold and 3-fold water respectively. 4 times the amount of elution, collecting eluent. Conclusion The process is simple and feasible, the separation effect is good, suitable for continuous industrial production of cistanche glycosides.
