目的研究hTERT mRNA及GGT mRNA-H在小肝癌患者外周血中的表达,并探讨其对肝癌早期诊断的价值。方法采用RT-PCR方法检测30例小肝癌患者、18例肝炎肝硬变患者以及12例正常受试者外周血hTERT mRNA及GGT mRNA-H的表达。结果小肝癌组患者hTERT mRNA及GGT mRNA-H表达阳性率分别为80.0%(24/30)和46.7%(14/30),均高于AFP的检测阳性率(26.7%,8/30),P<0.05。正常对照组均未检测到hTERT mRNA和GGT mRNA-H的表达。肝炎肝硬变组患者hTERT mRNA的表达阳性率为33.3%(6/18),未检测到GGT mRNA-H的表达。联合检测小肝癌患者hTERT mRNA及GGT mRNA-H表达的阳性率明显高于AFP的阳性率(86.7%比26.7%),P<0.05。结论 hTERT mRNA及GGT mRNA-H在小肝癌患者中特异性表达,联合检测外周血hTERT mRNA及GGT mRNA-H可提高小肝癌的早期诊断率。
Objective To investigate the expression of hTERT mRNA and GGT mRNA-H in peripheral blood of patients with small hepatocellular carcinoma (HCC) and to explore their value in the early diagnosis of HCC. Methods RT-PCR was used to detect the expression of hTERT mRNA and GGT mRNA-H in 30 patients with small hepatocellular carcinoma, 18 patients with cirrhosis and 12 normal subjects. Results The positive rates of hTERT mRNA and GGT mRNA-H in patients with small HCC were 80.0% (24/30) and 46.7% (14/30), respectively, which were higher than that of AFP (26.7%, 8/30) P <0.05. No expression of hTERT mRNA and GGT mRNA-H were detected in normal control group. The positive expression rate of hTERT mRNA in patients with hepatitis cirrhosis was 33.3% (6/18), and the expression of GGT mRNA-H was not detected. The positive rate of hTERT mRNA and GGT mRNA-H in combined detection of small hepatocellular carcinoma was significantly higher than that of AFP (86.7% vs. 26.7%, P <0.05). Conclusion hTERT mRNA and GGT mRNA-H are specifically expressed in small hepatocellular carcinoma patients. Combined detection of hTERT mRNA and GGT mRNA-H in peripheral blood can improve the early diagnosis rate of small hepatocellular carcinoma.
