目的 探讨武汉地区肾综合征出血热重型患者感染的汉坦病毒M片段G1基因变异的情况。方法 用RT PCR对 34份肾综合征出血热患者发病 1周内血清中的汉坦病毒G1基因扩增 ,并选取 7份重型 ,1份轻型及 1份中型患者的PCR产物测序 ,分别与标准株HV114和R2 2株相应序列比较。结果 1.Ⅰ型重型患者G1区在 15 1~ 2 10位核苷酸有意义突变的频率很高 ,而轻型患者则无此改变。 2 .Ⅱ型重型患者G1区 5 49位 ,5 5 2位和 6 92位核苷酸变异引起编码氨基酸的改变相同 ,中型患者无此特点。结论 这些核苷酸变异可能与病毒毒力增强有关 ,从而引起临床重症表现。
Objective To investigate the mutation of G1 gene of Hantavirus M fragment in severe patients with hemorrhagic fever with renal syndrome in Wuhan. Methods RT-PCR was used to amplify the Hantaan virus G1 gene in 34 patients with hemorrhagic fever with renal syndrome (HFRS) within one week after the onset of HFRS. The PCR products of 7 heavy, 1 light and 1 medium-sized patients were sequenced and compared with standard The corresponding sequences of strains HV114 and R2 2 were compared. Results 1. In type I patients with severe type G1, there was a high frequency of meaningful mutations in nucleotides 151-1 210, while no changes were found in those with mild type I diabetes. 2. Type II heavy patients in G1 area 5 49, 5 5 2 and 6 92 nucleotide mutations caused by coding amino acid changes the same, no such characteristics of medium-sized patients. Conclusion These nucleotide variations may be related to the enhanced virulence of the virus, leading to clinical severe manifestations.
