目的研究哺乳期多溴联苯醚-153(BDE-153)暴露对成年大鼠海马神经细胞凋亡及p53基因和蛋白表达的影响。方法将40只4日龄清洁级SD雄性大鼠按体重随机分成4组,分别为对照(橄榄油)组和1、5、10 mg/kg BDE-153染毒组,每组10只。于出生第10天(PND10),采用腹腔一次性注射方式进行染毒,染毒容量为10 ml/kg。染毒2个月后,测定海马神经细胞的凋亡率和TUNEL阳性细胞率及海马p53基因和蛋白的表达。结果与对照组相比,10 mg/kg BDE-153染毒组大鼠海马神经细胞TUNEL阳性细胞率和p53基因的表达均升高,5、10 mg/kg BDE-153染毒组大鼠海马神经细胞凋亡率和p53蛋白的阳性率均增高,差异有统计学意义(P<0.05)。且随着BDE-153染毒剂量的升高,大鼠海马神经细胞凋亡率和TUNEL阳性细胞率及p53基因的表达和p53蛋白的阳性率均呈上升趋势。结论 p53基因和蛋白可能参与了BDE-153诱导的神经细胞凋亡过程。
Objective To investigate the effects of lactational polybrominated diphenyl ether-153 (BDE-153) exposure on neuronal apoptosis and p53 gene and protein expression in the hippocampus of adult rats. Methods 40 male SD rats of 4-day-old clean grade were randomly divided into 4 groups according to body weight. They were control (olive oil) group and 1,5,10 mg / kg BDE-153 exposure group, 10 rats in each group. On the tenth day of birth (PND10), the rats were injected intraperitoneally with a single injection. The exposure volume was 10 ml / kg. After 2 months of exposure, the apoptotic rate of hippocampal neurons and the percentage of TUNEL positive cells and the expression of p53 gene and protein in hippocampus were determined. Results Compared with the control group, the TUNEL positive rate and the p53 gene expression in hippocampal neurons of 10 mg / kg BDE-153 group were significantly increased, while hippocampus of 5,10 mg / kg BDE-153 group Neuronal apoptosis rate and p53 protein positive rate were increased, the difference was statistically significant (P <0.05). The apoptotic rate of hippocampal neurons, the percentage of TUNEL-positive cells, the expression of p53 gene and the positive rate of p53 protein in rat hippocampus increased with the increase of BDE-153 dose. Conclusion p53 gene and protein may be involved in the neuronal apoptosis induced by BDE-153.
