目的检测系统性红斑狼疮(systemic lupus erythematosus,SLE)患者外周血及血清中CD200和多种自身抗体的表达水平,并探讨其相互关系。方法采用流式细胞术检测受试者外周血细胞表面CD200的表达,应用ELISA方法检测血清游离CD200水平,同时用印迹法检测患者相关自身抗体的阳性率。结果 SLE患者CD3+CD4+T细胞CD200的表达及血清游离CD200水平均显著高于健康对照者(P<0.05);SLE患者抗ds-DNA、Sm、ARPA、抗核小体抗体阳性率明显高于对照组;SLE患者自身抗体表达阳性者,其外周血及血清中CD200的表达显著高于自身抗体表达阴性者。结论 SLE患者CD200表达异常,且与SLE相关自身抗体的表达密切相关。
Objective To detect the expression levels of CD200 and autoantibodies in peripheral blood and serum of patients with systemic lupus erythematosus (SLE) and to explore their relationship. Methods Flow cytometry was used to detect the expression of CD200 on the surface of peripheral blood mononuclear cells. Serum free CD200 level was measured by ELISA. Meanwhile, the positive rate of autoantibodies was detected by Western blotting. Results The expression of CD200 and serum free CD200 in CD3 + CD4 + T cells of SLE patients were significantly higher than those in healthy controls (P <0.05). The positive rates of anti-dsDNA, Sm, ARPA and anti-nucleosome antibodies in SLE patients were significantly higher In the control group, the expression of CD200 in peripheral blood and serum of SLE patients with autoantibodies was significantly higher than those with negative autoantibodies. Conclusion The expression of CD200 in patients with SLE is abnormal and closely related to the expression of SLE autoantibodies.
