水稻扭曲叶突变体rtl1是利用甲基磺酸乙酯(EMS)诱变粳稻品种日本晴获得的。在苗期,该突变体的叶片就表现出皱缩和扭曲状。将该突变体分别与籼稻品种台中本地1号和浙辐802进行配组。遗传分析表明该突变体性状受1对隐性单基因控制。通过混合分离法,找到了位于第4染色体上的紧密连锁SSR标记RM1155,通过新发展的多态性STS标记,最终将该基因定位在STS标记T1591和SSR标记RM1359之间,其遗传距离分别为0.48和0.96cM。为进一步克隆该基因打下了基础。
Mutant rtl1, a mutant of rice, was obtained by inducting the Japonica rice variety Nipponbare with ethyl methanesulfonate (EMS). At the seedling stage, the leaves of the mutant appeared shriveled and twisted. The mutant was mixed with indica variety Taichung Local No.1 and Zhefu 802 respectively. Genetic analysis showed that the trait was controlled by a single recessive single gene. A tightly linked SSR marker RM1155 located on chromosome 4 was found by hybrid isolation method. The gene was finally located between the STS marker T1591 and the SSR marker RM1359 through the newly developed polymorphic STS marker. The genetic distances were 0.48 and 0.96 cM. Lay the foundation for further cloning this gene.
