为了改进奶牛布鲁氏菌PCR检出率低的问题,试验合成了一对PCR鉴定引物,通过对样品增菌培养来进行布鲁氏菌的PCR检测。结果表明:该方法能有效检出布鲁氏菌,灵敏度达1cfu,特异性好;通过对奶牛阴道分泌物24h增菌培养后,布鲁氏菌的检出率大大提高。说明结合细菌学检测的PCR检测方法能有效检测奶牛布鲁氏菌,提高了检测的灵敏度和布鲁氏菌病诊断的准确性。
In order to improve the detection rate of Brucella brucellosis in dairy cows, a pair of PCR identification primers were synthesized and PCR detection of Brucella was carried out by enriching the samples. The results showed that the method could detect Brucella effectively and had a sensitivity of 1 cfu with good specificity. The detection rate of brucellosis increased greatly after the vaginal secretions of cows were cultured for 24h. This shows that the combination of bacteriological detection of PCR detection method can effectively detect dairy cows Brucella, improve the detection sensitivity and brucellosis diagnosis accuracy.
