目的:检测结直肠癌患者外周血循环中黑素瘤抗原编码基因(melanoma antigen encoding gene,MAGE)-A3、MAGE-A6和MAGE-A10的表达情况,并分析其临床意义。方法:采用实时荧光定量PCR(real-time fluorescent quantitative PCR,RFQ-PCR)检测35例结直肠癌患者和15例正常对照者外周血中MAGE-A3、MAGE-A6和MAGE-A10mRNA的表达水平,并分析其与结直肠癌患者临床病理特征(性别、年龄、肿瘤部位、组织学分级、肿瘤浸润深度、淋巴结转移和Dukes分期)之间的关系。结果:所有外周血样本中均未检测出MAGE-A3mRNA的表达。结直肠癌组MAGE-A6mRNA表达阳性率显著高于正常对照组(P=0.0000),男性患者的表达水平高于女性患者(P=0.0459),淋巴结转移患者的表达水平也高于无转移患者(P=0.0000);结直肠癌组MAGE-A10mRNA表达阳性率也显著高于正常对照组(P=0.0224);淋巴结转移患者的表达水平也高于无转移患者(P=0.0000)。结论:结直肠癌患者外周血循环中MAGE-A6和MAGE-A10mRNA的表达在结直肠癌转移检测中具有重要意义,可能成为结直肠癌微转移诊断、治疗选择和预后判断中的重要临床靶标。
Objective: To detect the expression of melanoma antigen encoding gene (MAGE) -A3, MAGE-A6 and MAGE-A10 in peripheral blood of patients with colorectal cancer and analyze its clinical significance. Methods: The expression of MAGE-A3, MAGE-A6 and MAGE-A10 mRNA in 35 patients with colorectal cancer and 15 normal controls were detected by real-time fluorescent quantitative PCR (RFQ-PCR) And analyzed its relationship with clinicopathological features (gender, age, tumor location, histological grade, depth of tumor invasion, lymph node metastasis and Dukes stage) in patients with colorectal cancer. Results: No expression of MAGE-A3 mRNA was detected in all peripheral blood samples. The positive rate of MAGE-A6 mRNA in colorectal cancer was significantly higher than that in the normal control group (P = 0.0000). The expression level of MAGE-A6 mRNA in colorectal cancer patients was higher than that in female patients (P = 0.0459) P = 0.0000). The positive rate of MAGE-A10 mRNA in colorectal cancer was significantly higher than that in the normal control group (P = 0.0224). The expression level of MAGE-A10 mRNA in patients with lymph node metastasis was also higher than those without metastasis (P = 0.0000). Conclusion: The expression of MAGE-A6 and MAGE-A10 mRNA in peripheral blood of patients with colorectal cancer is of great significance in the detection of colorectal cancer metastasis and may be an important clinical target in the diagnosis, treatment choice and prognosis of colorectal cancer.
