目的研究拉西地平对异丙肾上腺素(Iso)、佛司可林和美兰(MB)增强心肌细胞收缩、舒张功能的影响。方法采用IonOptix单细胞动缘检测系统,测定心肌细胞收缩幅度和收缩、舒张速度等,指标均由计算机自动实时采集并记录。结果Iso、Fors-kolin和MB增强单个心肌细胞收缩幅度(ph)和单个心肌细胞收缩幅度/单个心肌细胞长度百分比(ph/bl),最大缩短速率(+dL/dt)和最大复长速率(-dL/dt),上述指标的增强均有显著性差异(P<0.01)。与对照组比较,Iso(10.0nmol/L)能使ph、ph/bl、+dL/dt和-dL/dt分别增加40%、41%、60%和50%。Forskolin(1.0μmol/L)能使ph、ph/bl、+dL/dt和-dL/dt分别增加89%、120%、106%、86%;用MB(10μmol/L)后,ph、ph/bl、+dL/dt和-dL/dt分别为0.15±0.03μm、12%±4%、2.3±0.4μm/s和2.4±0.5μm/s,用药前后差异有统计学意义(P<0.01)。3.0μmol/L的Larcidipine能抑制Iso引起ph、ph/bl、+dL/dt和-dL/dt的升高(P均<0.05),Lar-cidipine能同时降低Forskolin和MB引起ph、ph/bl、+dL/dt和-dL/dt的增加。结论Iso、Forskolin和MB使心肌细胞收缩和舒张功能均增强;Larcidipine可抑制Iso、Forskolin和MB的作用。
Objective To investigate the effects of lacidipine on the contraction and relaxation of cardiomyocytes induced by isoprenaline (Iso), forskolin and melanin (MB). Methods Using IonOptix single cell mobile edge detection system, the contractile amplitude, systolic and diastolic velocities of cardiomyocytes were measured. The indexes were collected and recorded automatically by the computer in real time. Results Iso, Fors-kolin and MB enhanced the contractile amplitude (ph) and single myocardial cell contraction amplitude / single myocardial cell length percentage (ph / bl), maximal shortening rate (dL / dt) and maximal growth rate -dL / dt), the above indicators were significantly different (P <0.01). Iso (10.0 nmol / L) increased ph, ph / bl, + dL / dt and -dL / dt by 40%, 41%, 60% and 50%, respectively, compared with the control. Forskolin (1.0μmol / L) increased ph, ph / bl, + dL / dt and -dL / dt by 89%, 120%, 106% and 86% / bl, + dL / dt and -dL / dt were 0.15 ± 0.03μm, 12% ± 4%, 2.3 ± 0.4μm / s and 2.4 ± 0.5μm / s, respectively ). Larcidipine at 3.0μmol / L could inhibit the increase of ph, ph / bl, + dL / dt and -dL / dt induced by iso (all P <0.05) , + DL / dt and -dL / dt increase. Conclusion Iso, Forskolin and MB enhanced the contractile and diastolic functions of cardiomyocytes. Larcidipine inhibited the effects of Iso, Forskolin and MB.
