目的观察蒺藜提取物对小鼠热损伤睾丸一氧化氮合酶(NOS)-一氧化氮(NO)系统的影响,探讨蒺藜对小鼠热损伤睾丸的保护修复作用机制。方法健康雄性小鼠30只,随机分为对照组、模型组和给药组,每组10只;建立睾丸热损伤模型,给予治疗剂量的蒺藜提取物;睾丸NOS活力和NO含量测定;睾丸NOS和caspase-3免疫组织化学染色,光学显微镜观察;酶联法检测血清中睾酮含量。结果模型组与对照组相比睾丸组织NOS活力和NO含量明显升高,睾丸i NOS阳性表达明显增加,睾丸caspase-3阳性细胞明显增多,血清睾酮含量明显降低。给药组与模型组相比睾丸组织NOS活力和NO含量明显降低,睾丸i NOS阳性表达明显减少,睾丸caspase-3阳性细胞明显减少,血清睾酮含量明显升高。结论蒺藜提取物通过调控NOS-NO系统,实现对小鼠热损伤睾丸的保护修复作用。
Objective To observe the effects of Tribulus terrestris extract on the nitric oxide synthase (NOS) - nitric oxide (NO) system in the testes of heat injury mice and to explore the protective mechanism of Tribulus terrestris on the heat injury of testes in mice. Methods Thirty healthy male mice were randomly divided into control group, model group and administration group, with 10 rats in each group. The model of testicular heat injury was established and the amount of Tribulus terrestris extract was administrated. The activity of testis NOS and content of NO were determined. And caspase-3 immunohistochemical staining, optical microscopy; enzyme-linked detection of serum testosterone levels. Results Compared with the control group, the activity of NOS and the content of NO in testis tissue of model group were significantly increased. The positive expression of iNOS in testis was significantly increased. The expression of caspase-3 positive cells in testis and serum testosterone were significantly decreased. Compared with the model group, the NOS activity and the content of NO in testis decreased significantly, the positive expression of i NOS in testis decreased significantly, the number of caspase-3 positive cells in testis decreased significantly, and the content of serum testosterone increased significantly. Conclusion Tribulus terrestris extract through the regulation of NOS-NO system, to achieve the protection of the repair of heat injury in mice testes.
