目的研究同时含人 p5 3、GM- CSF、B7- 1、IL- 2基因的重组腺病毒载体 Ad- m ultigenes,对肝癌细胞免疫原性的影响。方法应用人外周血淋巴细胞和肿瘤细胞的混合培养、外周血 T淋巴细胞杀伤活性试验 ,分析导入目的基因的人肝癌细胞免疫原性的变化。结果导入 Ad- m ultigenes的肝癌细胞系 Hep G2、BEL- 740 2 ,体外能刺激正常人外周血 T淋巴细胞的增殖 ,并能增强 T淋巴细胞的杀瘤细胞活性 ,加入 IL- 12有协同增强作用。结论肝癌细胞导入含 B7- 1基因的 Ad- m ultigenes后 ,其免疫原性得到增强
Objective To investigate the effect of recombinant adenovirus vector Ad-multigenes containing human p53, GM-CSF, B7-1 and IL-2 genes on the immunogenicity of hepatoma cells. Methods The mixed culture of human peripheral blood lymphocytes and tumor cells and the peripheral blood T lymphocyte killing activity test were used to analyze the changes in the immunogenicity of human hepatoma cells introduced with the target gene. Results Ad-m ultigenes hepatocellular carcinoma cell lines Hep G2 and BEL-740 2 were stimulated in vitro to stimulate the proliferation of normal human peripheral blood T lymphocytes and to enhance the activity of T lymphocyte killer cells. IL- 12 was added to enhance effect. Conclusion The immunogenicity of hepatoma cells was enhanced after the introduction of Ad- m ultigenes containing the B7-1 gene.
