目的探讨肝癌缺失基因1(DLC1)不同亚型对结肠癌细胞系增殖及迁移的抑制作用。方法通过qRT-PCR检测DLC1不同亚型在17对结肠癌组织与正常组织中的表达水平;同时过表达不同亚型的慢病毒感染结肠癌细胞SW1116,通过CCK8及Transwell实验观察不同亚型对结肠癌细胞株增殖及迁移的作用。结果 DLC1不同亚型在结肠癌组织中的表达均显著低于癌旁组织;过表达DLC1能够抑制结肠癌细胞系SW1116的增殖及迁移,其中DLC1亚型2(DLC1-isoform2)的作用效果最强。结论 DLC1的不同亚型在结肠癌组织中的表达均低于癌旁组织,DLC1-isoform2对抑制结肠癌细胞的增殖及迁移作用最强。
Objective To investigate the inhibitory effect of different subtypes of hepatocellular carcinoma deletion gene 1 (DLC1) on the proliferation and migration of colon cancer cell lines. Methods The expression of different DLC1 subtypes in 17 pairs of colon cancer tissues and normal tissues was detected by qRT-PCR. At the same time, different subtypes of lentivirus were overexpressed to express colon cancer cells SW1116. CCK8 and Transwell experiments were performed to evaluate the effect of different subtypes on colon The role of cancer cell proliferation and migration. Results The expression of DLC1 subtypes in colorectal cancer tissues was significantly lower than that in paracancerous tissues. Overexpression of DLC1 could inhibit the proliferation and migration of colon cancer cell line SW1116. DLC1-isoform2 was the most effective . Conclusion The expression of DLC1 in different subtypes of colon cancer tissues is lower than that of paracancerous tissues. DLC1-isoform2 has the strongest inhibitory effect on the proliferation and migration of colon cancer cells.
