目的探讨经验方清脉791-2冲剂对肢体缺血动物模型的促血管生成作用及其疗效机理,为临床应用提供理论基础。方法建立大鼠后肢严重缺血模型,以活血化瘀代表药物丹参作为对照药。造模及药物治疗后2、4周分别观测以下指标:X线血管造影片血管面积,病理组织切片碱性磷酸酶染色观察毛细血管面积及密度,免疫组化法观察血管内皮细胞生长因子(VEGF)及碱性成纤维细胞生长因子(bFGF)的光密度值。结果791-2可显著增加血管造影片侧枝血管面积,增加病理组织切片毛细血管面积及密度,一定程度地增强及延长VEGF的表达,对bFGF的作用相对较弱。结论清脉791-2对肢体缺血实验动物具有一定的促血管生成作用;促进VEGF及bFGF的表达,是其可能的作用机制。
Objective To investigate the proangiogenic effect of Fangqing Qingmai 791-2 Granule on animal model of limb ischemia and its curative effect mechanism, and provide theoretical basis for clinical application. Methods A model of severe hindlimb ischemia in rats was established. The blood and blood stasis were used as the reference drug. After modeling and drug treatment, the following indicators were observed at 2 and 4 weeks: X-ray angiogram vascular area, pathological tissue section alkaline phosphatase staining observed capillary area and density, immunohistochemical observation of vascular endothelial cell growth factor (VEGF ) and the optical density of basic fibroblast growth factor (bFGF). Results 791-2 can significantly increase the collateral vascular area of angiograms, increase the capillary area and density of pathological tissue sections, and enhance and prolong VEGF expression to a certain extent. The effect on bFGF is relatively weak. Conclusion Qingmai 791-2 has a certain angiogenic effect on experimental animals with limb ischemia; promoting the expression of VEGF and bFGF is its possible mechanism.
