目的探讨恒古骨伤愈合剂对兔骨折愈合的影响。方法 2016年6—8月于浙江中医药大学动物实验研究中心选取普通级健康雄性兔12只,适应性饲养3 d后,随机分成治疗组与对照组,各6例。两组兔均复制骨折模型,治疗组灌服恒古骨伤愈合剂,对照组灌服等量0.9%氯化钠溶液,两组均连续给药5周。比较两组兔治疗前后空腹血糖(FPG)、三酰甘油(TG)、体质量,治疗1周、3周、5周后骨形态发生蛋白2(BMP-2)、转化生长因子β1(TGF-β1)、血小板衍生因子(PDGF)、碱性成纤维细胞生长因子(b FGF)水平。结果治疗前后两组兔FPG、TG水平及体质量比较,差异无统计学意义(P>0.05)。治疗1周后,治疗组TGF-β1水平高于对照组(P<0.05);治疗1周后,两组BMP-2、PDGF、b FGF水平比较,差异无统计学意义(P>0.05);治疗3周后,两组BMP-2、TGF-β1、PDGF、b FGF水平比较,差异无统计学意义(P>0.05);治疗5周后,治疗组BMP-2、TGF-β1、PDGF、b FGF水平高于对照组(P<0.05)。结论恒古骨伤愈合剂可提高骨折兔BMP-2、TGF-β1、PDGF、b FGF水平,有利于兔骨折愈合。
Objective To investigate the effect of Henggu bone healing agent on fracture healing in rabbits. Methods From June to August of 2016, 12 healthy male rabbits were selected from Animal Experimental Research Center of Zhejiang University of Chinese Medicine. After 3 days of adaptive feeding, they were randomly divided into treatment group and control group, with 6 in each. The fracture models were duplicated in both groups. The treatment group was fed with the anti-aging bone healing agent, while the control group was given 0.9% sodium chloride solution. The two groups were given continuous administration for 5 weeks. The levels of fasting blood glucose (FPG), triglyceride (TG), body weight, bone morphogenetic protein 2 (BMP-2) and transforming growth factor β1 (TGF- β1), platelet-derived factor (PDGF) and basic fibroblast growth factor (b FGF). Results Before and after treatment, there was no significant difference in FPG and TG levels and body weight between the two groups (P> 0.05). One week after treatment, the level of TGF-β1 in the treatment group was higher than that in the control group (P <0.05). There was no significant difference in the levels of BMP-2, PDGF and b FGF between the two groups after one week of treatment (P> 0.05). After 3 weeks of treatment, the levels of BMP-2, TGF-β1, PDGF and b FGF in the two groups had no significant difference (P> 0.05) b FGF levels were higher than the control group (P <0.05). CONCLUSION: Healing agents can increase the levels of BMP-2, TGF-β1, PDGF and b FGF in rabbits, which is in favor of rabbit fracture healing.
