目的 :探讨芩夏止哮颗粒对卵蛋白 (OVA)致敏哮喘小鼠T细胞活化和细胞因子水平的影响。方法 :用卵蛋白致敏和喷雾攻击复制小鼠过敏性气道炎症模型 ,观察芩夏止哮颗粒对脾细胞增殖反应和mIL 2R表达 ,以及肺泡灌洗液 (BALF)和脾细胞培养上清中IL 4、IL 5和IFN γ水平的影响。结果 :正常小鼠脾细胞自发增殖反应 ,在ConA或OVA刺激下增殖反应 ,以及脾细胞mIL 2R表达均较模型对照小鼠低 (P <0 .0 1)。芩夏止哮颗粒能明显抑制哮喘小鼠脾细胞自发增殖反应和在ConA及OVA刺激下增殖反应及IL 2R的表达 (P <0 .0 1)。模型对照小鼠肺泡灌洗液 (BALF)和脾细胞培养上清中IL 4、IL 5含量显著升高 (P <0 .0 1) ,而IFN γ模型对照脾细胞培养上清中水平低于正常小鼠。芩夏止哮颗粒则能显著降低BALF中和脾细胞分泌IL 4、IL 5的水平 ,提高IFN γ产生水平 (P <0 .0 1或P <0 .0 5 )。结论 :芩夏止哮颗粒治疗哮喘机制与其能抑制哮喘小鼠体内T淋巴细胞的活化 ,促进Th1/Th2类细胞因子产生平衡有关
Objective: To investigate the effects of Qixia Xieer Granules on T cell activation and cytokine levels in asthmatic mice sensitized with ovalbumin (OVA). METHODS: Mouse allergic airway inflammation models were replicated with ovalbumin and spray challenge. The proliferation of spleen cells and expression of mIL 2R in Qixia Xiehao Granules were observed, as well as in culture supernatants of BALF and spleen cells. Effects of IL 4, IL 5 and IFN gamma levels. RESULTS: Spontaneous cell proliferation in normal mice, proliferative response to stimulation with ConA or OVA, and expression of mIL 2R in spleen cells were lower than those in model control mice (P < 0.01). Qixia Xie-erh granules could significantly inhibit the spontaneous proliferation of asthmatic mice spleen cells and proliferative response and IL 2R expression under ConA and OVA stimulation (P < 0.01). The levels of IL 4 and IL 5 in the BALF and splenocyte culture supernatants of the model control mice were significantly increased (P < 0.01), while those in the IFN-γ model control spleen cell culture supernatants were lower than those in the model control mice. Normal mice. Qixiazheheng granules can significantly reduce the levels of IL-4 and IL5 secreted by BALF and spleen cells and increase the level of IFNγ production (P <0.01 or P <0.05). Conclusion: Qixia Xiehao Granules can reduce the activation of T lymphocytes in asthmatic mice and promote the balance of Th1/Th2 cytokines in the treatment of asthma.
