背景:目前有关醒脑静注射液(xingnaojinginjection,XNJI)对脑缺血再灌注损伤(cerebralischemia-reperfusioninjury,CIRI)时血清白细胞介素8水平的影响,及其脑功能保护机制研究尚少。目的:观察XNJI对CIRI家兔血清白细胞介素8水平的影响,并探讨其脑保护机制。设计:完全随机设计,对照实验研究。地点与材料:本研究的地点为温州医学院病理生理教研室。材料为选用28只日本大耳白兔,雌雄不拘,体质量2.2~3.0kg,由温州医学院实验动物中心提供。干预:28只家兔随机分为假手术对照组、缺血再灌注组和醒脑静注射液组,分别在缺血前、缺血30min、再灌注30min,60min,120min取静脉血,测定血清白细胞介素8浓度,实验结束取脑组织观察脑超微结构的变化。主要观察指标:血清白细胞介素8浓度的动态变化;脑组织超微结构的改变。结果:脑缺血再灌注期间,缺血再灌注组不同时点血清白细胞介素8水平明显高于醒脑静注射液组犤缺血30min为(0.62±0.18)ng/L及(0.43±0.08)ng/L,P<0.05;再灌注30min,为(0.73±0.19)ng/L及(0.45±0.09)ng/L;再灌注60min,为(0.87±0.29)ng/L及(0.47±0.06)ng/L;再灌注120min为(1.03±0.43)ng/L及(0.44±0.07)ng/L,再灌注各时点组均P<0.01犦,超微结构发生异常改变;醒脑静注射液组不同时点血清白细胞介素8浓度
BACKGROUND: At present, there are few studies on the effects of Xingnaojing injection (XNJI) on serum interleukin-8 level during cerebral ischemia-reperfusion injury (CIRI) and its protective mechanism of brain function. OBJECTIVE: To observe the effect of XNJI on serum interleukin-8 levels in CIRI rabbits and to explore its brain protection mechanism. Design: Completely random design, controlled experimental study. Location and Materials: The location of this study was the Department of Pathophysiology, Wenzhou Medical College. The material was 28 Japanese white rabbits, either male or female, weighing 2.2 to 3.0 kg, provided by the Experimental Animal Center of Wenzhou Medical College. Intervention: 28 rabbits were randomly divided into sham operation control group, ischemia reperfusion group and Xingnaojing injection group. Venous blood was taken before ischemia, 30 minutes of ischemia, 30 minutes of reperfusion, 60 minutes, and 120 minutes of reperfusion. At the end of the experiment, the brain tissue was taken to observe the changes of brain ultrastructure. MAIN OUTCOME MEASURES: Dynamic changes of serum interleukin-8 concentration; changes in brain ultrastructure.
