目的:探讨SATB1与前列腺癌增殖及侵袭发生的关系。方法:体外培养前列腺癌细胞系(LNCa P、PC3、DU145),MMT法检测其增殖能力;Tanswell细胞侵袭实验检测其侵袭能力;RT-PCR检测其SATB1 mRNA的表达情况;Western blot检测其SATB1蛋白质的表达。结果:前列腺癌细胞PC3和DU145的增值能力及侵袭能力比LNCaP强(P<0.01),PC3和DU145细胞中SATB1 mRNA及蛋白质表达水平均显著高于LNCaP细胞(P<0.01)。结论:SATB1在前列腺癌表达水平可能与前列腺癌的增殖和侵袭正相关。
Objective: To investigate the relationship between SATB1 and proliferation and invasion of prostate cancer. Methods: Prostate cancer cell lines (LNCa P, PC3 and DU145) were cultured in vitro. The proliferation of prostate cancer cells was detected by MMT assay. The invasion ability was detected by Tanswell cell invasion assay. The expression of SATB1 mRNA was detected by RT-PCR. expression. Results: The proliferation and invasion ability of prostate cancer cells PC3 and DU145 were stronger than that of LNCaP (P <0.01). The expression of SATB1 mRNA and protein in PC3 and DU145 cells was significantly higher than that in LNCaP cells (P <0.01). Conclusion: The expression of SATB1 in prostate cancer may be positively correlated with the proliferation and invasion of prostate cancer.
