应用骨髓细胞长期培养方法动态观察10例AML贴壁细胞生长特点,探讨AML基质细胞在白血病发生、发展中的作用。发现5/10例贴壁层较早形成完全融合。培养2~3周各例AML贴壁层出现一些大型多角扁平细胞,其中2例M5出现一些大型巨噬细胞;培养至8~9周,6/10例AML融合消失,贴壁层仅留下一种大型多角扁平细胞和大量由其介导获独立生长的造血细胞(为幼稚细胞),表明这些大型基质细胞功能较为活跃。我们认为基质细胞组成的改变可能是影响造血细胞增殖、分化方向的原因,参与白血病的发生、发展。
The long-term culture of bone marrow cells was used to dynamically observe the growth characteristics of 10 AML adherent cells, and to explore the role of AML stromal cells in the occurrence and development of leukemia. It was found that 5/10 cases of the adherent layer formed an early fusion. In the 2 to 3 weeks of culture, some large polygonal flat cells appeared in each AML adherent layer, of which 2 macrophages appeared in M5. In 8 to 9 weeks of culture, 6/10 cases of AML fusion disappeared, and the adherent layer remained only. A large number of horny flat cells and a large number of hematopoietic cells (which are naive cells) mediated by their independent growth indicate that these large stromal cells are more active. We believe that changes in the composition of stromal cells may be responsible for the direction of hematopoietic cell proliferation and differentiation and participate in the occurrence and development of leukemia.
