目的:探讨核因子(NF)Ⅰ对血小板源性生长因子(PDGF)增强皮肤成纤维细胞转化生长因子β(TGF-β)信号通路的影响作用.方法:构建NF Ⅰ-C基因慢病毒载体并且确定慢病毒转染成纤维细胞最佳条件,采用Western印迹法测定PDGF对经NF Ⅰ-C基因病毒转染后成纤维细胞的TGF-β1及其受体Ⅱ(TβRⅡ)蛋白表达的影响作用.结果:①NF Ⅰ-C慢病毒载体转染皮肤成纤维细胞的最佳转染值为50;②皮肤成纤维细胞经NF Ⅰ-C慢病毒转染后NF Ⅰ-C蛋白的表达较非病毒转染细胞显著升高(P<0.05);③正常皮肤成纤维细胞与PDGF共同培养48 h后TGF-β1和TβRⅡ蛋白表达较空白对照显著升高(P<0.05);④皮肤成纤维细胞经NF Ⅰ-C慢病毒转染后与PDGF共同培养后48 h后TGF-β1和TβRⅡ蛋白表达均较正常对照皮肤成纤维细胞和空病毒转染皮肤成纤维细胞显著降低(P<0.05).结论:血小板源性生长因子可增强皮肤成纤维细胞TGF-β1和TGF-β受体Ⅱ的表达,而NF Ⅰ-C表达增加可抑制PDGF增强TβRⅡ表达的作用.“,”Objective:To assess the effect of nuclear tactor Ⅰ-C(NFⅠ-C) on transforming growth factor(TGF-β) signal pathway induced by PDGF in fibroblasts.Methods:The lentiviral vector of NFⅠ-C was constructed,and the optimal condition of lentiviral infection was established in fibroblasts.Western blotting was used to assess the effect of PDGF on expression levels of TGF-β1 and its receptor(TβR Ⅱ) in fibroblasts transfected with NFⅠ-C.Results:(1) The optimal multiplicity of NFⅠ-C lentiviral infection was 50;(2) Expression levels of NFⅠ-C protein was significantly increased in fibroblasts transfected with NFⅠ-C lentiviral vector compared with that transfected with blank NFⅠ-C lentiviral vector(P<0.05);(3) Expression levels of TβR Ⅱ and TGF-β1 were also significantly increased in normal fibroblasts treated with PDGF for 48h compared with that in blank control group(P<0.05);(4) Expression levels of TβRⅡand TGF-β1 were significantly decreased in fibroblasts transfected with NFⅠ-C lentiviral vector compared with that in both normal fibroblasts and those treated with blank NFⅠ-C lentiviral vector(P<0.05).Conclusions:PDGF can increase the expression of TβR Ⅱ and TGF-β1.Elevated expression levels of NFⅠ-C can inhibit PDGF-induced increase in TβRⅡand TGF-β1 expression.
