目的:观察补脾强力复方对实验性自身免疫性重症肌无力(EAMG)模型大鼠胸腺组织中糖皮质激素受体(GR)mRNA表达的影响。方法:将Lewis大鼠按体重随机分为6组,即:正常组、模型组、强的松组(5.4 mg/kg)、补脾强力复方不同剂量组(剂量分别为4.75 g/kg,7.12 g/kg,9.49 g/kg),除正常组外,均采用鼠源性AchR-α亚基97-116肽段序列(R97-116)免疫接种复制EAMG模型。药物治疗后,ELISA法检测各组大鼠的AchR-Ab、皮质醇水平,RT-Q-PCR技术检测大鼠胸腺组织GR mRNA表达水平。结果:与正常组比较,模型组GR mRNA相对表达量降低(P<0.01),与模型组比较,西药组、中药大、中、小剂量组大鼠AchR-Ab含量下降(P<0.05),强的松组、中药大剂量组和中剂量组GR mRNA相对表达量增加(P<0.05)。结论:补脾强力复方可提高胸腺组织GR mRNA的表达,这可能是其治疗MG的作用机制之一。
OBJECTIVE: To observe the effect of BuBuQiangLiFang on glucocorticoid receptor (GR) mRNA expression in thymus of experimental autoimmune myasthenia gravis (EAMG) model rats. Methods: Lewis rats were randomly divided into 6 groups according to body weight: normal group, model group, prednisone group (5.4 mg / kg) and different doses of Bupi Qiangdu Compound (4.75 g / kg, 7.12 g / kg, 9.49 g / kg). All the mice except the normal group were immunized with the murine AchR-α subunit 97-116 peptide sequence (R97-116) for immunoprecipitation. After drug treatment, the AchR-Ab and cortisol levels in each group were detected by ELISA, and the GR mRNA expression in thymus tissue was detected by RT-Q-PCR. Results: Compared with the normal group, the expression of GR mRNA in the model group decreased (P <0.01). Compared with the model group, the content of AchR-Ab in the large, medium and small dose groups decreased significantly (P <0.05) The relative mRNA level of GR mRNA in prednisone group, high-dose group and middle-dose group increased (P <0.05). Conclusion: BBS can increase the expression of GR mRNA in thymus tissue, which may be one of the mechanisms of its treatment of MG.
