目的:研究丹参多酚酸盐对HepG2215细胞X蛋白(HBx)表达的影响。方法:将HepG2215细胞分为对照组和药物组,药物组设三个浓度(100μg/ml2、00μg/ml4、00μg/ml)。待药物作用细胞24h后,荧光定量PCR法检测HepG2215细胞HBx mR-NA表达水平,ELISA法检测细胞内HBx蛋白的表达水平。结果:与对照组相比,药物组在不同的剂量100μg/ml2、00μg/ml、400μg/ml下均可明显降低HBx mRNA的表达水平,但不呈现剂量-反应关系。ELISA结果显示:药物组在不同的剂量100μg/ml2、00μg/ml、400μg/ml下均可明显降低胞内HBx蛋白表达的水平,且呈现剂量-反应关系。结论:丹参多酚酸盐可显著影响HepG2215细胞中X蛋白的表达。
Objective: To study the effect of salvianolate on the expression of X protein (HBx) in HepG2215 cells. Methods: HepG2215 cells were divided into control group and drug group. The drug group consisted of three concentrations (100μg / ml2,00μg / ml4,00μg / ml). After treated with drugs for 24 hours, the expression of HBx mR-NA in HepG2215 cells was detected by real-time quantitative PCR, and the expression of HBx protein was detected by ELISA. Results: Compared with the control group, the drug group could significantly reduce the expression of HBx mRNA at different doses of 100μg / ml, 2μg / ml and 400μg / ml, but did not show the dose-response relationship. The results of ELISA showed that the drug group could significantly decrease the intracellular HBx protein expression at dose of 100μg / ml, 2μg / ml and 400μg / ml, and showed a dose-response relationship. Conclusion: salvianolate can significantly affect the expression of X protein in HepG2215 cells.
