目的探讨火把花根对大鼠油酸0.04 mL/mg致急性肺损伤(ALI)的保护作用。方法 48只雄性Wistar大鼠,随机分为3组:对照组(C组,n=12)、ALI组(A组,n=18)和火把花根预防组(H 组,n=18)。A组尾静脉注射油酸0.04 ml/kg建立ALI模型,H组以火把花根片600 mg·kg-1·d-1连续灌胃10 d,第10灌胃后1 h注射油酸,C组注射生理盐水0.04ml/kg。于注射油酸后4h处死大鼠,处死大鼠前即刻采左心血,用流式细胞术测定中性粒细胞(PMN)、单核细胞、淋巴细胞表面CD11a、CD11b 和CD18的表达水平,测定肺通透指数(LPI)、支气管肺泡灌洗液(BALF)中白细胞(WBC)计数和活化 PMN百分比。结果油酸可诱导LPI、BALF中WBC和活化PMN百分比升高及WBC表面CD11a、 CD11b和CD18表达上调,火把花根预先给药可减弱上述改变(P<0.05)。结论火把花根预先给药对大鼠油酸致ALI有一定的保护作用,可能与下调WBC表面CD11a/CD18和CD11b/CD18的表达有关。
Objective To investigate the protective effect of the roots of Hemonia on acute lung injury (ALI) induced by 0.04 mL/mg oleic acid in rats. Methods Forty-eight male Wistar rats were randomly divided into three groups: control group (group C, n=12), ALI group (group A, n=18), and flower bud root prevention group (group H, n=18). The ALI model was established by tail vein injection of oleic acid 0.04 ml/kg in group A. In group H, the root tablets of Rhizoma curcuma was treated with 600 mg·kg-1·d-1 for 10 days, and oleic acid was given 1 h after the 10th intragastric injection. In group C, normal saline 0.04 ml/kg was injected. Rats were sacrificed 4 h after the injection of oleic acid. Left heart blood was collected immediately before the rats were killed. Flow cytometry was used to measure the expression levels of PMN, monocytes, and lymphocytes on the surface of CD11a, CD11b, and CD18. Lung permeation index (LPI), white blood cell (WBC) counts in bronchoalveolar lavage fluid (BALF) and percentage of activated PMN. RESULTS: Oleic acid induced LPI, increased percentage of WBC and activated PMN in BALF, and increased expression of CD11a, CD11b and CD18 on WBC surface. The pre-administration of H. praecox could weaken the above changes (P<0.05). Conclusion The pre-administration of H. fruticosa has certain protective effects on ALI induced by oleic acid, which may be related to the down-regulation of the expression of CD11a/CD18 and CD11b/CD18 on WBC surface.
