目的:观察顺铂是否能够逆转鼻咽癌紫杉醇耐药细胞HNE-2/taxol和5-8F/taxol的耐药性。方法:运用集落形成实验观察不同浓度顺铂对鼻咽癌亲本细胞(HNE-2及5-8F)和紫杉醇耐药细胞(HNE-2/taxol及5-8F/taxol)的生长抑制率,并且判断经低浓度顺铂预处理后,耐药细胞耐药指数的改变。通过等效剂量和联用指数分析,评价紫杉醇和顺铂联合用药的效果。运用FCM法检测不同浓度顺铂处理后鼻咽癌亲本细胞和耐药细胞的凋亡率变化。结果:两种鼻咽癌紫杉醇耐药细胞株对顺铂的敏感性均显著高于其亲本细胞(P<0.05)。顺铂与紫杉醇合用时,对鼻咽癌亲本细胞的增殖抑制能够产生相加作用,而对耐药细胞的增殖抑制则能够产生协同作用。经过低浓度顺铂预处理后,耐药细胞的耐药指数明显降低。在顺铂作用下,鼻咽癌紫杉醇耐药细胞的早期凋亡率显著高于亲本细胞(P<0.05)。结论:顺铂可以逆转鼻咽癌细胞的紫杉醇耐药性。顺铂和紫杉醇联合作用于鼻咽癌紫杉醇耐药细胞,能够产生协同效应。
AIM: To investigate whether cisplatin can reverse the drug resistance of paclitaxel-resistant cell line HNE-2 / taxol and 5-8F / taxol. Methods: The colony formation assay was used to observe the growth inhibitory rates of cisplatin on the growth of NPC cells (HNE-2 and 5-8F) and paclitaxel resistant cells (HNE-2 / taxol and 5-8F / taxol) To determine the low concentration of cisplatin pretreatment, drug resistant cell resistance index changes. The effect of paclitaxel and cisplatin combination therapy was evaluated by equivalent dose and combination index analysis. The apoptosis rate of nasopharyngeal carcinoma parental cells and drug-resistant cells after different concentrations of cisplatin treatment were detected by FCM. Results: The sensitivity of paclitaxel-resistant cell lines to cisplatin was significantly higher than that of their parental cells (P <0.05). The combination of cisplatin and paclitaxel can inhibit the proliferation of nasopharyngeal carcinoma parental cells and inhibit the proliferation of drug-resistant cells. After low concentrations of cisplatin pretreatment, resistance index of drug-resistant cells was significantly reduced. In cisplatin, the early apoptotic rate of paclitaxel-resistant cells in nasopharyngeal carcinoma was significantly higher than that of the parental cells (P <0.05). Conclusion: Cisplatin can reverse paclitaxel resistance in nasopharyngeal carcinoma cells. Combination of cisplatin and paclitaxel in paclitaxel-resistant cells of nasopharyngeal carcinoma cells can produce synergistic effects.
