生肌玉红膏通过下调TGF-β1/Smads抑制人增生性瘢痕成纤维细胞增殖和胶原分泌

目的探讨生肌玉红膏(SJYHG)对人增生性瘢痕成纤维细胞(h HSF)增殖、胶原分泌及TGF-β1/Smads通路的影响。方法采用原代培养的h HSF模型,加入SJYHG培养后,CCK-8法测定450nm波长各孔溶液吸光度值(A值),荧光定量PCR检测α-SMA、I、III型胶原的mRNA表达,WB检测α-SMA、I、III型胶原和P-Smad2、P-Smad3的蛋白表达。结果 SJYHG浓度依赖地极显著地抑制h HSF增殖(P<0.001),并显著抑制了α-SMA和I、III型胶原的mRNA和蛋白表达(P<0.05),显著下调P-Smad2和P-Smad3的蛋白表达(P<0.001)。结论 SJYHG具有潜在的抑制增生性瘢痕的作用。 Objective To investigate the effects of SJYHG on proliferation, collagen secretion and TGF-β1 / Smads pathway in human hypertrophic scar fibroblasts (h HSF). Methods Primary culture h HSF model was used. After SJYHG culture, the absorbance value (A value) of each well of 450nm wavelength was determined by CCK-8 method. The mRNA expression of α-SMA, type I and type III collagen was detected by fluorescence quantitative PCR The protein expression of α-SMA, type I and type III collagen and P-Smad2 and P-Smad3 were detected. Results SJYHG significantly inhibited h HSF proliferation (P <0.001) in a concentration-dependent manner and significantly inhibited the mRNA and protein expression of α-SMA and collagen type I and III (P <0.05), significantly down-regulated the expressions of P-Smad2 and P- Smad3 protein expression (P <0.001). Conclusion SJYHG has a potential role in inhibiting hypertrophic scars.
