应用高速逆流色谱分离纯化了苦豆子中的生物碱。以氯仿-甲醇-0.08mol/LKH2PO4(27:20:13,v/v/v)为溶剂体系,以上相为固定相,下相为流动相,分离得到纯度分别为96.37%及98.10%的金雀花碱及槐定碱。以氯仿-甲醇-0.08mol/LKH2PO4(27:20:21,v/v/v,盐溶液pH6.50)为溶剂体系,分离得到纯度为92.53%的氧化苦参碱。分离所得组分用标准品进行对照,用反相高压液相色谱进行分析。该法有效改善了溶剂体系的两相乳化情况,具有简便、快速、产物纯度高的优点。
The alkaloids in Sophora alopecuroides were isolated and purified by high speed countercurrent chromatography. The mobile phase consisted of chloroform-methanol-0.08 mol / L KH2PO4 (27:20:13, v / v / v) as the mobile phase. The phases were the stationary phase and the lower phase was the mobile phase. The purity of 96.37% and 98.10% Sparine and sophoridine. Using chloroform-methanol-0.08mol / L KH2PO4 (27:20:21, v / v / v, salt solution pH6.50) as solvent system, oxymatrine with purity of 92.53% was isolated. The separated components were compared with standards and analyzed by reversed-phase high pressure liquid chromatography. The method effectively improves the two-phase emulsification of the solvent system, has the advantages of simplicity, rapidness and high product purity.
