目的:研究硒酸酯多糖与紫杉醇单独及序贯给药对体外培养乳腺癌MCF-7细胞的生长抑制作用及对细胞周期的影响。方法:采用MTT法及SRB法检测药物对乳腺癌MCF-7细胞体外杀伤作用,应用金氏公式判断联合用药效果,流式细胞仪检测细胞周期及凋亡率。结果:MTT及SRB法结果显示,药物对MCF-7细胞的抑制作用呈剂量依赖性,两者联合作用效果优于单独作用组,30mg/L的硒酸酯多糖与0.5~2nmol/L紫杉醇联合应用表现为协同作用,与4nmol/L紫杉醇联合应用表现为相加作用。流式细胞术结果显示,紫杉醇作用于G1期,同时添加组及序贯给药组既可发生S期和G2/M期周期阻滞,又可诱导凋亡,但凋亡率低于Taxol单独组。结论:紫杉醇与硒酸酯多糖对乳腺癌MCF-7细胞有协同抑制增殖和诱导凋亡的作用。
OBJECTIVE: To study the inhibitory effects of selenomethionate polysaccharide and paclitaxel alone and sequentially on the growth of MCF-7 breast cancer cells and on the cell cycle. Methods: MTT assay and SRB method were used to detect the cytotoxicity of MCF-7 on MCF-7 cells in vitro. The effect of combination therapy was evaluated by Kim’s formula. The cell cycle and apoptosis rate were determined by flow cytometry. Results: The results of MTT and SRB showed that the drug inhibited MCF-7 cells in a dose-dependent manner, and the combined effect was better than that of the single treatment group. Selenium 30mg / L and 0.5 ~ 2nmol / L paclitaxel combined Application showed synergy, combined with 4nmol / L paclitaxel showed an additive effect. Flow cytometry showed that paclitaxel in the G1 phase, while adding the group and sequential administration of both S and G2 / M cycle arrest, but also induce apoptosis, but the apoptosis rate was lower than that of Taxol alone group. Conclusion: Paclitaxel and selenate polysaccharide have the synergistic effect on inhibiting proliferation and inducing apoptosis of breast cancer MCF-7 cells.
