[目的]观测四君子汤对脾气虚模大鼠血清5-HIAA、MTL表达影响。[方法]使用随机平行对照方法,30只(6~8周)健康SD大鼠适应性饲养1周,随机数字表法分为3组,空白组、脾气虚组、四君子汤组,10只/组。模型复制:实验第5d,用力竭游泳和饮食不节的复合因素建立脾气虚模型。造模成功后,给予四君子汤灌胃治疗6d;四君子汤灌胃期间,空白组和模型组灌等量蒸馏水形成对照。实验第24d(造模14d),各组大鼠均于末次给药干预处理后禁食不禁水24h,10%水合氯醛腹腔麻醉(3mL/kg),剖腹,腹主动脉取血,4℃放置1h,3000转离心分离10min,分离使用不含热原和内毒素的试管,操作过程避免任何细胞刺激,收集,置于-20℃冰箱中待用。ELISA法观测5-HIAA、MTL水平、一般状态。[结果]血清5-HIAA平均含量脾气虚组、四君子组均高于白组(P<0.01);四君子组低于脾气虚组(P<0.01);血清MTL平均含量脾气虚组、四君子组均低于空白组(P<0.01);四君子组高于脾气虚组(P<0.01)。[结论]四君子汤灌胃干预脾气虚大鼠,可抑制5-HIAA表达,升高IL-6表达,有利于促进胃排空、缩短胃排空时间、增强食欲、增加进食量,进而改善脾气虚大鼠食欲下降、摄食减少、体重减轻等症状,起到一定调控作用。
[Objective] To observe the effect of Sijunzi Decoction on the serum 5-HIAA and MTL expression in asthenia model rats. [Methods] Thirty healthy (6-8 weeks) healthy SD rats were reared for 1 week with randomized parallel control. The rats were randomly divided into three groups: blank group, spleen deficiency group, Sijunzi Decoction group, group. Model duplication: On the 5th day of experiment, the model of spleen-qi deficiency was established by the combined factors of exhaustive swimming and diet. After the model was established successfully, Sijunzi decoction was given to the stomach for 6 days. During the perfusion of Sijunzi Decoction, the rats in the blank group and the model group were infused with the same amount of distilled water as control. On the 24th day of experiment (model 14d), rats in each group were fasted for 24 h, 10% chloral hydrate intraperitoneally (3 mL / kg), cesarean section, abdominal aorta blood and 4 ℃ Placed 1h, centrifuge 3000 rpm 10min, the separation of the use of pyrogen-free endotoxin tubes, the operation to avoid any cell stimulation, collected, placed in -20 ℃ refrigerator stand-by. [Results] The average contents of 5-HIAA in serum were lower than those in the spleen-qi deficiency group and the Sijunzi group (P <0.01), those in the Sijunzi group were lower than those in the spleen-Qi deficiency group (P <0.01) Lower than the blank group (P <0.01); Sijunzi group was higher than the spleen Qi group (P <0.01). [Conclusion] Sijunzi decoction can inhibit 5-HIAA expression and increase IL-6 expression by intragastric administration of Sijunzi Decoction in the stomach, which is beneficial to promote gastric emptying, shorten the time of gastric emptying, increase appetite, increase the food intake, and then improve the temper Asthenia rats decreased appetite, reduced feeding, weight loss and other symptoms, play a role in regulation.
