目的 :本研究拟探讨人参皂苷Rg1对Aβ2 5 - 35诱导的BT32 5细胞株脑啡肽酶表达的影响。方法 :应用MTT比色法检测一定剂量Aβ2 5 - 35 (2 5 μmol/L)和不同浓度人参皂苷Rg1(5、 10、 2 0 μmol/L)对BT32 5细胞存活率的影响 ,应用RT -PCR观察细胞中NEP基因表达的变化。结果 :2 5 μmol/L的Aβ2 5 - 35作用于BT32 5细胞株 ,BT32 5细胞存活率下降 ,细胞内NEP的表达降低。人参皂苷Rg1能提高Aβ2 5 - 35处理的BT32 5细胞存活率 ,使细胞内NEP表达增高。结论 :一定剂量的Aβ2 5 - 35能造成细胞损伤和细胞内NEP表达降低的模型 ,人参皂苷Rg1对Aβ2 5 - 35造成的细胞毒性具有拮抗作用 ,增高细胞内NEP的表达。
Objective: This study was to investigate the effect of ginsenoside Rg1 on the enkephalinase expression induced by Aβ25-35 in BT32 5 cell line. METHODS: MTT colorimetric assay was used to determine the effects of doses of Aβ25-35 (25 μmol/L) and different concentrations of ginsenoside Rg1 (5, 10, 20 μmol/L) on the viability of BT32 5 cells. The change of NEP gene expression in cells was observed by PCR. Results: 25 μmol/L Aβ25-35 acted on the BT32 5 cell line, the survival rate of BT32 5 cells decreased, and the expression of intracellular NEP decreased. Ginsenoside Rg1 can increase the survival rate of BT32 5 cells treated with Aβ2 5-35 and increase the expression of NEP in cells. CONCLUSION: A certain dose of Aβ2 5-35 can cause cell damage and a decrease in intracellular NEP expression. Ginsenoside Rg1 can antagonize the cytotoxicity of Aβ25-35 and increase the expression of NEP in cells.
